Gene therapy: Editing out genetic damage

Jan. 31, 2013 ? New design guidelines from researchers in Singapore simplify the development of targeted therapies for muscular dystrophy and other diseases.

The dystrophin protein offers critical support to muscle fibers. Mutations affecting dystrophin's expression cause the muscle-wasting disease muscular dystrophy. In Duchenne muscular dystrophy (DMD), these mutations take the form of small sequence changes that make much of the dystrophin gene (DMD) untranslatable, yielding nonfunctional protein or no protein at all.

Therapies based on a strategy known as 'exon skipping' could undo the damage from these mutations. Development of such treatments is set to accelerate, thanks to research by a team led by Keng Boon Wee of the A*STAR Institute of High Performance Computing and Zacharias Pramono of the National Skin Centre in Singapore.

Proteins are translated from messenger RNA transcripts of genes; however, only certain RNA regions -- known as exons -- actually encode protein, and these are enzymatically spliced together prior to translation. Several clinical studies have demonstrated that small 'antisense oligonucleotide' (AON) molecules that bind mutated DMD exons can induce elimination of those defective exons during splicing, yielding shorter but largely functional versions of dystrophin. "We are cautiously optimistic that AON-induced exon skipping could be the first effective therapy for DMD patients," says Wee.

Unfortunately, DMD arises from many different mutations, and targeted AON design remains a time-consuming, trial-and-error process. To address this challenge, Wee and Pramono sought to define the characteristics of AONs that efficiently promote exon-skipping. They used computational analysis to zoom in on exonic sequences that coordinate splicing. They also identified regions of suitable length within dystrophin RNA transcripts that span these sequences and would be accessible to AONs in living cells.

The researchers thus derived a set of guidelines enabling them to effectively design AONs that targeted nine different exons affected in DMD patients. For each exon, at least one AON proved capable of boosting dystrophin expression to clinically relevant thresholds in cultured muscle cells (see image). "Our proposed set of factors resulted in a reasonable success rate of designing efficient AONs -- 61% versus 38% using semi-empirical methods," says Wee. Clinical studies have already demonstrated the promise of efficient exon skipping in treating DMD patients.

Wee notes that other diseases arising from abnormal RNA processing could also benefit from this approach. However, his team is also exploring this method as a general strategy to abort production of disease-causing proteins in cancer and other conditions. "In contrast to small-molecule inhibitor drugs that can target only about 10% of the human genome, this approach could downregulate most human genes," Wee says.

Iran's New Indigenous Qaher 313' - Business Insider

Iran yesterday revealed its most recent indigenous fighter jet ? the ?Qaher 313?.?The prototype Q-313 was presented to?President Mahmoud Ahmadinejad, as a ceremonial gift during the country's Ten-Day Dawn celebration. The festivities commemorate the 1979 revolution when the U.S. embassy in Tehran taken by force.?

In the previous days, the Iranian Defense Minister Brigadier General Ahmad Vahidi had said, ?The aircraft will be different from the other fighter jets Iran has already made.?

Indeed, based on the first photographs released by the FARS News Agency, the new stealthy jet has a peculiar design. It features hard edges and those distinctive edges and angle of the U.S.?F-22?and the twin tail shape much similar to that of the?F-35 Lightning II.

The Q-313 has large, seemingly fixed canards, and little wings whose external section is canted downward, while the?canopy material is at least odd (based on its transparency, it looks like Plexiglas or something like that).

The cockpit seems basic (a bit too much for a modern plane ? note the lack of wiring?behind the front panel).

Putting aside the Qaher's implausible aerodynamics and Hollywood sheen, last week's announcement of a coming new domestic fighter was not insignificant given the Iranian air force has no more than a few dozen strike aircraft.

Most of those it inherited from the prior regime during the 1979 revolution. That regime change, involving a huge U.S. hostage situation, made it far more difficult to maintain the country's then-new fleet of American F-14s.

Tehran has a domestically built fighter already. At least a handful have been sighted during air shows and displays, but this new model is a drastic change in design and ability.

Of course, Iran is famous for building models and systems it never intends to fly, but if this is an actual prototype it may be designed to fill a uniquely Iranian requirement.

Air dominance against the nearly 200 aircraft of the U.S. Fifth Fleet out of Bahrain isn't a realistic goal for Tehran. What it would look to do in any open engagement with the U.S. would be to overwhelm American surface ships with small, high-speed boats, and missiles of its own.?

Those boats would immediately be set upon by a fleet of U.S. helicopters and fixed wing aircraft. Most naval experts are quick to point out that a fight between a helo' and a boat generally isn't much of a fight at all. Un-distracted helicopters have immense advantages, but once they start taking fire or need to react to other airborne attacks, that changes.

Perhaps Iran isn't imagining a fleet of stealthy fighter/bombers, based on some design similar to this new display, but a few dozen single seat craft with an array of missiles, a couple guns, and the ability to fly low and slow over the Gulf.

A string of planes to compliment the one military project they've been working hardest over the last decade, but rarely ever mention. That plane would be a handy addition to cover the?Bradstone based speed boat, they believe can take on the Fifth Fleet and bring down the U.S. Navy.

While Iran does have a few HESA Shahed light attack/recon helicopters, they're

?modestly armed and have a limited range of a couple hundred miles.

Iran's other domestically produced jet, the Saeqeh fighter?is?designed to engage other fighters in mid-air combat also not a practical choice against the might of the U.S. Fleet.

A Qaher variation against the Bradstone would fill a niche, but is obviously pure speculation. But speculating has some value when examining Iran's military projects because like most countries, the weapons Tehran speaks of the least seem to mean the most.?

This couldn't be more true than in the case of the?ill-gotten Bradstone and Iran's blossoming missile inventory that's about as extensive and diverse as it is deadly and proven.

MEXICO CITY (AP) ? Rescuers searched for survivors and authorities promised a thorough investigation after an office building blast killed 25 people and injured 101 at the headquarters of Mexico's state-owned oil company, Petroleos Mexicanos.

The cause of the basement explosion in an administrative building next to the iconic, 51-story Pemex tower in Mexico City remained a mystery early Friday, with President Enrique Pena Nieto urging people not to speculate. Theories ranged from an electrical fire to an air conditioning problem to a possible attack.

"We have no conclusive report on the reason," Pena Nieto told reporters. "We will work to get to the bottom of the investigation to find out, first, what happened ... and if there are people responsible in this case, that we apply the full weight of the law against them."

Some 46 people remained hospitalized after the Thursday afternoon blast, some gravely injured and others with cuts, fractures and burns. Authorities said the dead were 17 women and eight men.

More than 500 firefighters, soldiers and rescue workers dug through chunks of concrete with dogs, trucks and a Pemex crane.

Interior Minister Miguel Osorio Chong said it was uncertain if any of the roughly 10,000 people who work in the five-building headquarters were still trapped, but that the search would continue. The explosion occurred at about 3:45 p.m., just as the administrative shift was about to end. It hit the basement and first two floors, which rescuers said all collapsed onto each other.

"There is a lot of risk," rescuer German Vazquez Garcia said of working on the site.

Pemex first said it had evacuated the tower and 14-story administrative building because of a problem with the electrical system. The company later tweeted that the Attorney General's Office was investigating the explosion.

Ana Vargas Palacio was distraught as she searched for her missing husband, Daniel Garcia Garcia, 36, who works in the building where the explosion occurred. She said she last talked to him a couple hours earlier.

"I called his phone many times, but a young man answered and told me he found the phone in the debris," Vargas said. The two have an 11-year-old daughter. His mother, Gloria Garcia Castaneda, collapsed on a friend's arm, crying "My son. My son."

Gabriela Espinoza, 50, a Pemex secretary for 29 years, was on the second floor of the tower when she said she heard two loud explosions and a third smaller one.

"There was a very loud roar. It was very ugly," she said.

Espinoza's co-worker, Tomas Rivera, 32, worked on the ground floor where the explosion occurred and said the force knocked him to the basement, fracturing his wrist and jaw. The injured were taken to two Pemex hospitals and other facilities, including the Red Cross hospital in the Polanco neighborhood near the oil company's office headquarters, where relatives huddled in the waiting room for news of their loved ones. Some walked out of meetings with the hospital social worker joyous, while others came out crying.

"We were talking and all of sudden we heard an explosion with white smoke and glass falling from the windows," said Maria Concepcion Andrade, 42, who lives on the same block as the Pemex building. "People started running from the building covered in dust. A lot of pieces were flying."

Streets surrounding the building were closed as evacuees wandered around, and rescue crews loaded the injured into ambulances.

Pemex, created as a state-owned company in 1938, has nearly 150,000 employees and in 2011 produced about 2.5 million barrels of crude oil a day, according to its website, with $111 billion in sales. Pena Nieto, who took office in December, has made Pemex reform the center of his platform, with a plan to pump new investment into a company whose profits feed much of Mexico's federal budget, but which has fallen behind other oil companies in production, technology and exploration.

Shortly before the explosion, Operations Director Carlos Murrieta reported via Twitter that the company had reduced its accident rate in recent years. Most Pemex accidents have occurred at pipeline and refinery installations.

A fire at a pipeline metering center in northeast Mexico near the Texas border killed 30 workers in September, the largest-single toll in at least a decade for the company.
